The Antiepileptic Effect of Zonisamide(P-132)


Abstract
Zonisamide, new antiepiletic drug, was administered to 25 epileptic patients(13 refractory and 12 new cases) for 6 months. The drug efficacy, serum level of zonisamide and side effects was investigated. Clinically improved or disappeared of seizures were 16 in 25 patients(8 in 13 of refractory cases and 8 in 12 of new cases). In type of seizures, rate of improved was 66.7% in generalized tonic clonic sezure, 88.8% in generalized tonic clonic sezure with partial onset, 25.0% in complex partial seizure, 100% in simple partial seizure. Neverthless mixed type with complex partial seizure and generlized tonic clonic seizure was not responded to the zonisamide. The average effective serum concentration of zonisamide was 21.8% ug/ml and mean dose given was 4.8mg/kg/day(23.4 ug/ml in refractory case and 20.3ug/ml new case). Side effects were observed in 13(52.0%) of 25 cases. These were most commonly loss of weight(32.0%) and anorexia(28.0%).These side effects were frequently found within three months after initiation of administraed and disappeared thereafter.
